低频静电场辐射对酿酒酵母生长代谢的影响

Effects of low frequency electrostatic field radiation on growth and metabolism of Saccharomyces cerevisiae

  • 摘要: 为了探究低频静电场辐射对酿酒酵母生长代谢的影响和作用机理。本实验运用60 Hz低频静电场对酿酒酵母进行辐射处理,通过扫描电子显微镜、流式细胞仪、酶标仪和紫外分光光度计等仪器对酿酒酵母的生长趋势、形态特征、葡萄糖代谢、胞内蛋白总量和胞内乙醇脱氢酶的活性进行测定分析。结果表明:低频静电场辐射条件下生长的酿酒酵母缩短了生长迟滞期,加快了其对数生长期,且其生长速率相比自然生长提高了14.5%。辐射条件下生长的酿酒酵母对葡萄糖的消耗量在时间为16 h时比自然生长提高了15.64%,其胞内乙醇脱氢酶(ADH)的比活力比自然生长提高了7.25%,但胞内蛋白的总量无显著变化。因此,低频静电场辐射可影响酿酒酵母原有的代谢机制,对其生长代谢过程具有明显的促进效应。
    Abstract: In order to explore the effect and mechanism of low frequency electrostatic field radiation on the growth and metabolism of Saccharomyces cerevisiae,Saccharomyces cerevisiae was radiated by 60 Hz low frequency electrostatic field,and its growth curve,morphological characteristics,dextrose metabolism,total intracellular protein and the enzyme activity of alcohol dehydrogenase were investigated by scanning electron microscopy,flow cytometry,microplate reader and ultraviolet spectrophotometer in this work. Experimental results showed that low frequency electrostatic field radiation could shorten the growth delay period of Saccharomyces cerevisiae and accelerate its logarithmic growth period,its growth rate was 14.5% higher compared with the natural growth. In addition,the consumption of dextrose of the radiation growth Saccharomyces cerevisiae (LFEF treatment for 16 h)was increased by 15.64%,and the specific activity of intracellular alcohol dehydrogenase(ADH)was 7.25% higher than that of natural growth,but the total intracellular protein had no significantly different. It was indicated that the low frequency electrostatic field radiation can affect the original metabolic mechanism of Saccharomyces cerevisiae and promote its growth and metabolism process.
